J Strength Cond Res 31(5): 1221–1228, 2017—One of the unique features of an exercise is that it leads to a simultaneous increase of antagonistic mediators. On the one hand, exercise elevates catabolic proinflammatory cytokines. On the other hand, exercise stimulates anabolic components such as heat shock proteins (HSPs), which protect against stressors. Therefore, the study was designed to evaluate the blood level of HSP27 and its relationship with muscle damage and inflammatory mediators in elite Greco-Roman wrestlers during training periods differed in type and intensity exercise. Ten male wrestlers (21.2 ± 2.1 years) were observed during the conditioning camps at preseason (January), at the beginning of tournament season (April), and during tournament season (June). Twelve healthy and untrained men (19.2 ± 0.4 years) were considered a reference group. The serum levels of inflammatory mediators and HSP27 in wrestlers were significantly different from nonathletes. In wrestlers, reactive oxygen and nitrogen species H 2 O 2 , NO, and 3-nitro, cytokines interleukin-1β and tumor necrosis factor α, and also HSP27 reached the highest levels at preseason (January) or tournament season (June) when the special training predominated (>30% training load) over directed training (approximately 10% training load). Creatine kinase activity also demonstrated the highest level during the same training periods (January 2,315 ± 806 IU·L −1 ; June 3,139 ± 975 IU·L −1 ). The regression analysis revealed the relationship of HSP27 level with muscle damage ( r s = −0.613, p < 0.001), and also with inflammatory mediators. The results of this study show that wrestling training modulates HSP27 level, which is significantly related with skeletal muscle damage and inflammatory response, and suggest that measure of HSP27 level can be useful diagnostic tool in biochemical assessment of athletes to increase their performance. J Strength Cond Res 36(3): 615–623, 2022—We examined the effects of high-intensity functional training (HIFT) on cardiorespiratory and neuromuscular performance, as well as on inflammatory and muscle damage markers. Thirteen physically active healthy volunteers (aged 28.3 ± 3.8 years, 5 men and 8 women) underwent 8 weeks of a group HIFT program performed 3 times per week. Each session consisted of 4 rounds of a 9-exercise circuit (30-second exercise and 15-second recovery). During the first and last weeks of training, venous blood was sampled daily to monitor changes in serum C-reactive protein (CRP) and creatine kinase (CK). After 8 weeks of HIFT, body fat decreased by 0.64 ± 1.01 kg ( p = 0.041), maximal oxygen uptake improved by 1.9 ± 2.2 ml·kg −1 ·min −1 ( p = 0.009), countermovement jump by 2.6 ± 1.5 cm ( p = 0.001), bench press 1-repetition maximum (1RM) by 4.5 ± 3.8 kg ( p = 0.001), maximum number of bench press repetitions at 65% 1RM by 4 ± 5 repetitions ( p = 0.03), and abdominal muscle endurance by 6 ± 4 repetitions ( p < 0.001). In both week 1 and week 8 of training, CK increased mildly in the morning after the first session of the week (main effect for day, p = 0.008), whereas no significant changes were observed in CRP ( p = 0.31). During week 8, CK on all days was ∼32% lower compared with week 1 (160 vs. 235 U·L −1 ; main effect of week 1 vs. week 8, p = 0.027), whereas CRP remained unchanged ( p = 0.225). This HIFT program was effective in improving cardiorespiratory and neuromuscular physical fitness without causing significant inflammation or muscle damage in physically active subjects. Abstract Biological processes linked to intramuscular inflammation during myogenous temporomandibular disorder (TMDM) are largely unknown. We mimicked this inflammation by intra-masseteric muscle (MM) injections of complete Freund’s adjuvant (CFA) or collagenase type 2 (Col), which emulates tissue damage. CFA triggered mechanical hypersensitivity at 1d post-injection was mainly linked to processes controlling chemotactic activity of monocytes and neutrophils. At 5d post-CFA, when hypersensitivity was resolved, there was minimal inflammation whereas tissue repair processes were pronounced. Low dose Col (0.2U) also produced acute orofacial hypersensitivity that was linked to tissue repair, but not inflammatory processes. High dose Col (10U) triggered prolonged orofacial hypersensitivity with inflammatory processes dominating at 1d post-injection. At pre-resolution time point (6d), tissue repair processes were underway and a significant increase in pro-inflammatory gene expressions compared to 1d post-injection were detected. RNA-seq and flow cytometry showed that immune processes in MM were linked to accumulation of macrophages, natural killer and natural killer T cells, dendritic cells and T-cells. Altogether, CFA and Col treatments induced different immune processes in MM. Importantly, orofacial hypersensitivity resolution was preceded with repairs of muscle cell and extracellular matrix, an elevation in immune system gene expression and accumulation of distinct immune cells in MM.

Background and Study Aim. The aim of this study is to investigate the effects of different types of contractions on muscle damage and muscle fatigue in sedentary individuals. Material and Methods. Thirty healthy male sedentary individuals participated in the study. Strength training in different types of contractions applied in the study was applied 3 times a week for 8 weeks. Before the study, the training loads were determined by making maximal force measurements of all subjects. The 30 subjects participating in the study were divided into 3 groups: isometric (n = 10), concentric (n = 10) and eccentric (n = 10) contraction group. Appropriate amount of blood samples was taken from the elbow vein 2 times from all subjects, before the studies and at the end of the 8-week strength training. Results. It was observed that eight-week strength training did not cause muscle fatigue in all groups and did not create a statistically significant difference (P> 0.05). Strength training with isometric and concentric contractions for eight weeks significantly increased serum lactate dehydrogenase (LDH), C-reactive protein (CRP), myoglobin (Mb), interleukin 6 (IL-6) levels, while concentric strength training significantly reduced serum aspartate amino transferase (AST) levels. Strength training with eccentric contractions significantly increased serum LDH, CRP, AST, Mb and IL-6 levels, while significantly reducing serum tumor necrosis factor alpha (TNF-α) levels. Strength training with eccentric contractions significantly increased serum creatine kinase (CK), CRP, AST, IL-6 and Mb levels compared to strength training with isometric and concentric contractions at the end of the eight-week study period, but did not show the same significant effect in other parameters.
