Peer-reviewed literature and traditional medicine sources report that black seed (Nigella sativa) oil and its active constituents possess pharmacological properties supporting their use in treating various conditions, including different types of cancer, asthma, and neurological disorders.
Although the seeds of cumin (Cuminum cyminum L.) are widely used as a spice for their distinctive
aroma, they are also commonly used in traditional medicine to treat a variety of diseases. The
literature presents ample evidence for the biomedical activities of cumin, which have generally
been ascribed to its bioactive constituents such as terpenes, phenols, and flavonoids. Those health
effects of cumin seeds that are experimentally validated are discussed in this review. Black seeds
(Nigella sativa), which are totally unrelated to C. cyminum, have nevertheless taken the name
‘Black cumin’ and used in traditional systems of medicine for many disorders. Numerous preclinical
and clinical trials have investigated its efficacy using the seed oil, essential oil, and its
main constituent thymoquinone (TQ). These investigations support its use either independently or
as an adjunct along with conventional drugs in respiratory problems, allergic rhinitis, dyspepsia,
metabolic syndrome, diabetes mellitus, inflammatory diseases, and different types of human
cancer. Multiple studies made in the last decades validate its health beneficial effects particularly
in diabetes, dyslipidemia, hypertension, respiratory disorders, inflammatory diseases, and cancer.
Nigella sativa seeds also possess immune stimulatory, gastroprotective, hepatoprotective,
nephroprotective, and neuroprotective activities. TQ is the most abundant constituent of volatile
oil of N. sativa seeds, and most of the medicinal properties of N. sativa are attributed mainly to TQ.
All the available evidence suggests that TQ should be developed as a novel drug in clinical trials.
BACKGROUND & OBJECTIVE
Medicinal plants have attracted great attention in the recent years and is increasingly applied instead of the chemical drugs. Several documents showed that herbal medicine traditionally and clinically applied in the cure and prevention of several diseases. In the recent years, different medicinal plants and their main components have been chosen in neurological therapy. The less toxic effects, availability, and lower price of medicinal plants versus synthetic substances make them as excellent and simple selection in the treatment of nervous diseases. Nigella sativa (N. Sativa) L. (Ranunculaceae), well recognized as black cumin, has been utilized as a medicinal plant that has a strong traditional background. Thymoquinone (TQ) is one of the main active components of the volatile oil of N. sativa seeds and most effects and actions of N. Sativa are mainly related to TQ. The several pharmacological properties of N. sativa and TQ have been found, for example; anti-tumor, anti-microbial, anti-histaminic, immunomodulatory, anti-inflammatory, and anti-oxidant effects. Many reviews have investigated this valuable plant and its components, but none of them focused on their neuroprotective effects. Therefore, the aim of the present review was to show comprehensive and neuropharmacological properties of N. sativa and TQ. In this review, various studies on scientific databases regarding the effects of N. sativa and TQ in neurological diseases have been introduced. Studies on the neuroprotective effects of N. sativa and TQ which were published between1979 and 2018, were searched using various databases. The results of these studies showed that N. sativa and TQ have the protective effects against neurodegenerative diseases, including; Alzheimer, depression, encephalomyelitis, epilepsy, ischemia, Parkinson, and traumatic brain injury have been discussed in the cell lines and experimental animal models. Although there are many studies indicating the beneficial actions of this plant in the nervous system, the number of research projects relating to the human reports is rare.
CONCLUSION
Therefore, better designed clinical trials in humans are needed to confirm these effects.
The current review investigates the effects of black seed (Nigella sativa) on human health, which is also used to encapsulate and oxidative stable in different food products. In recent decades, many extraction methods, such as cold pressing, supercritical fluid extraction, Soxhlet extraction, hydro distillation (HD) method, microwave-assisted extraction (MAE), ultrasound-assisted extraction, steam distillation, and accelerated solvent extraction (ASE) have been used to extract the oils from black seeds under optimal conditions. Black seed oil contains essential fatty acids, in which the major fatty acids are linoleic, oleic, and palmitic acids. The oxidative stability of black seed oil is very low, due to various environmental conditions or factors (temperature and light) affecting the stability. The oxidative stability of black seed oil has been increased by using encapsulation methods, including nanoprecipitation, ultra-sonication, spray-drying, nanoprecipitation, electrohydrodynamic, atomization, freeze-drying, a electrospray technique, and coaxial electrospraying. Black seed, oil, microcapsules, and their components have been used in various food processing, pharmaceutical, nutraceutical, and cosmetics industries as functional ingredients for multiple purposes. Black seed and oil contain thymoquinone as a major component, which has anti-oxidant, -diabetic, -inflammatory, -cancer, -viral, and -microbial properties, due to its phenolic compounds. Many clinical and experimental studies have indicated that the black seed and their by-products can be used to reduce the risk of cardiovascular diseases, chronic cancer, diabetes, oxidative stress, polycystic ovary syndrome, metabolic disorders, hypertension, asthma, and skin disorders. In this review, we are focusing on black seed oil composition and increasing the stability using different encapsulation methods. It is used in various food products to increase the human nutrition and health properties.
Abstract As a crucial organ, the lung is exposed to various harmful agents that may induce inflammation and oxidative stress, which may cause chronic or acute lung injury. Nigella sativa, also known as black seed, has been widely used to treat various diseases and is one of the most extensively researched medicinal plants. Thymoquinone (TQ) is the main component of black seed volatile oil and has been proven to have antioxidant, anti-inflammatory, and antineoplastic properties. The potential therapeutic properties of TQ against various pulmonary disorders have been studied in both in vitro and in vivo studies. Furthermore, the application of nanotechnology may increase drug solubility, cellular absorption, drug release (sustained or control), and drug delivery to lung tissue target sites. As a result, fabricating TQ as nanoparticles (NPs) is a potential therapeutic approach against a variety of lung diseases. In this current review, we summarize recent findings on the efficacy of TQ and its nanotypes in lung disorders caused by immunocompromised conditions such as cancer, diabetes, gastric ulcers, and other neurodegenerative diseases. It is concluded that TQ nanoparticles with anti-inflammatory, antioxidant, antiasthma, and antitumor activity may be safely applied to treat lung disorders. However, more research is required before TQ nanoparticles can be used as pharmaceutical preparations in human studies.
Mounting evidence support the potential benefits of functional foods or nutraceuticals for human health and diseases. Black cumin (Nigella sativa L.), a highly valued nutraceutical herb with a wide array of health benefits, has attracted growing interest from health-conscious individuals, the scientific community, and pharmaceutical industries. The pleiotropic pharmacological effects of black cumin, and its main bioactive component thymoquinone (TQ), have been manifested by their ability to attenuate oxidative stress and inflammation, and to promote immunity, cell survival, and energy metabolism, which underlie diverse health benefits, including protection against metabolic, cardiovascular, digestive, hepatic, renal, respiratory, reproductive, and neurological disorders, cancer, and so on. Furthermore, black cumin acts as an antidote, mitigating various toxicities and drug-induced side effects. Despite significant advances in pharmacological benefits, this miracle herb and its active components are still far from their clinical application. This review begins with highlighting the research trends in black cumin and revisiting phytochemical profiles. Subsequently, pharmacological attributes and health benefits of black cumin and TQ are critically reviewed. We overview molecular pharmacology to gain insight into the underlying mechanism of health benefits. Issues related to pharmacokinetic herb-drug interactions, drug delivery, and safety are also addressed. Identifying knowledge gaps, our current effort will direct future research to advance potential applications of black cumin and TQ in health and diseases.
